Timing for Multiple SPI reads

I am planning to use an STM32U5 SPI peripheral and I want to do a SPI read of a "black box" that doesnt exist yet. currently the planned output of this device has this configuration where the chip select is high for 5 bytes of data.

the STM32U5 is capable of reading SPI frames up to 32 bits. Can i read this in sections of 8 bits at a time with one long chip select? Ideally, using DMA to pass these 40 bit reads into memory.  If this would not work, then how would the chip select and CLK format need to change?
