I installed spc5studio that downloaded from ST website and it works correctly. But in software, I can't debug MCU with spc5studio. I checked windows/preferences/Spc5studio but there is not "Debug Options". How should I debug with spc5studio?

You need to install PLS UDE from https://www.pls-mc.com/service/downloads/download-spc5-udestk/
and then you can install Eclipse Plugins in SPC5Studio
